Wayne County was founded in 1796 and organized in 1815. It was the sixth county in the former Northwest Territory formed on August 15, 1796, from positions of territorial Knox County, territorial Hamilton County, and some uncategorized territories. Wayne County originally encompassed most of the Upper Peninsula, the entire area of the Lower Peninsula, and other smaller sections that are now part of Illinois, Indiana, northern Ohio, and Wisconsin.
On August 15, 1796, acting Governor and Territorial Secretary Winthrop Sargent declared that Wayne County boundaries were to begin at the mouth of the Cuyahoga River, then to the southernmost part of Lake Michigan. The border moved along the western shore north to Lake Superior and then along the territorial waters through Lake Huron, Lake Erie, and Lake St. Clair back to the starting point.
Wayne County was first divided into townships on November 1, 1798. The county was divided into four townships of Detroit, Mackinaw, Hamtramck, and Sergeant. At that time, the extent of Wayne County included the present state of Michigan. This is besides the parts of Ohio, Indiana, and Wisconsin.

Wayne County is named after General Anthony Wayne, General George Washington's most lethal and trusted soldier. Wayne was also nicknamed "Mad Anthony Wayne" for his bravery and courage. Anthony Wayne contributed significantly to the development of this county. Wayne County originally encompassed most of the Upper Peninsula and Lower Peninsula. This saw Wayne County become one of the largest counties in the 1700s.
